Haemophilus influenzae is a Gram negative and specific human pathogen that causes acute bacterial meningitis , septicemia , epiglotitis , bacterimia , otitis media . the goal of this study is one of the conserved proteins in this bacterium and designing of a domestic vaccine against it. ProteinP2 (38-40 KDa ) comprises more than 50 percent of all the proteins in outer membrane of Haemophilus and is expressesed by non-capsulated strains of H.influenzae type b.
This protein is indictaed as a proper candidate for vaccine to non-capsulated Haemophilus influenzae .in this study , epidemiology of ompP2 gene was surveyed using clinical samples isolated from 30 patients . the bacteria were cultured in chocolate agar mediaand Biochemical tests performed for them. After DNA purification , PCR method was performed for samples . all the samples were positive for ompP2 gene . after sequencing of 6 samples and comparison of ompP2 gene in all samples with domestic strains that exist in Gene Bank , similarities and differences due to nucleotide deletion , addition or translocation were defined by MEGA 4 software.
PCR products were digested by Alu1 and EcoR1 for defining of polymorphism patterns in given gene. Cutting patterns in 21 samples (98%-99%) were similar to domestic samples and 9 (91%-98%) of them were distinct. our results are indicated that ompP2 gene organization is similar to distinct strains of Haemophilus ifluenzae recorded in Gene Bank (accessionno: J3359.1, M93268.1, M93269.1, M93270.1) and polymorphism has occurred in cutting region by Alu1 enzyme. Thus this protein can be considered as a potential vaccine of Haemophilus in future studies.
